Minipig Midazolam Metabolism Correlated With Enzyme Abundance at Specific Developmental Stages
Enzyme abundance and metabolic activity provide related but different developmental measurements.
Enzyme abundance and metabolic activity provide related but different developmental measurements.
What the source reports
The laboratory study quantified liver CYP proteins across fetal, neonatal and adult Göttingen minipigs. It compared abundance with activity in the same biological samples. A relationship between CYP3A22 abundance and midazolam metabolism was observed at a specified early postnatal stage.
Limits and uncertainty
These were animal microsomal measurements rather than measured drug clearance in children. Correlation at one age does not establish an identical relationship throughout development. Species-specific enzymes, protein expression and experimental substrates restrict direct extrapolation.
Application to expert review
When explaining pediatric model inputs, distinguish measured protein, laboratory activity and predicted whole-body exposure. Human maturation cannot be inferred by directly scaling an animal age. The work supplies useful model-building data without validating an individual infant’s midazolam clearance or sensitivity.
