Dependence Review’s Receptor Adaptation Model Cannot Diagnose a Particular Patient

Biological explanations of tolerance are distinct from individual diagnostic evidence.

Biological explanations of tolerance are distinct from individual diagnostic evidence.

What the publication shows

The expert review described GABA-receptor adaptations and changes in excitatory signaling as proposed contributors to tolerance and physical dependence. It integrated molecular and clinical literature and discussed prevention and withdrawal management across several contexts.

Limits of the evidence

The synthesis did not measure receptor changes in every long-term user or provide a diagnostic biomarker for benzodiazepine use disorder. Mechanistic findings from different experimental systems cannot establish the timing or magnitude of adaptation in a particular person.

Application to expert review

For an individual opinion, use the actual exposure history, symptom pattern, function and clinical assessment. Increasing dose may have several explanations and does not independently prove one receptor mechanism. Keep physical adaptation, withdrawal phenomena and problematic-use behavior as related but separate evidentiary questions.
